Un aventurier célèbre, star de la télévision, est porté disparu lors d'un voyage au fin fond de l'Amazonie. Durant des années, accompagné de sa femme et de son fils, le Dr Emmet Cole a partagé - via son show télé - ses découvertes de contrées éloignées. Aujourd'hui, la chaîne finance une expédition filmée façon télé réalité pour tenter de le retrouver. Une équipe de cameramen suit de près les recherches menées entre autres par la famille et des amis du disparu. Tous embarquent à bord d'un bateau qui remonte l'Amazone et va les amener à faire des rencontres pour le moins effrayantes...
